Toxic effect of streptozotocin on cultured Schwann cells

Abstract
In order to evaluate the toxic effect of streptozotocin(STZ) on cultured mouse Schwann cells, the neurotoxicity was measured by NR assay after mouse Schwann cells were grown in various concentrations of STZ for 24 hours. In addition, the neuroprotective effect of IGF-¥± on the neurotoxicity induced by STZ was exxamined. The results were as follows : 1) NR50 value was a concentration of 50¥ìM STZ(highly toxic [NR50<100¥ìM, Borenfreund et al., 1988]), 2) STZ decreased markedly cell viability of cultured Schwann cells in a dose-dependent manner, 3) Cultured Schwann cells treated with 50¥ìM of STZ for 24 hours resulted in the degenerative changes of cells, vacuole, granulation and cytolysis, 4) Cell viability of cultured Schwann cells was increased remarkably after pretreatment of cells with 200 ng/§¢ IGF-¥± for 2 hours before exposure of STZ. From these results, it is suggested that STZ is very toxic in these cultures and IGF-¥± prevents cultured Schwann cells from the STZ-mediated neurotoxicity.
